sertraline + memantine

sertraline + memantine is a combination therapy consisting of the selective serotonin reuptake inhibitor (SSRI) sertraline and the NMDA receptor antagonist memantine. This combination is being investigated as an augmentation strategy for patients with obsessive-compulsive disorder (OCD) who have not achieved remission after initial sertraline monotherapy. Sertraline works by inhibiting the serotonin transporter (SERT), thereby increasing synaptic serotonin levels. Memantine acts as a low-to-moderate affinity, non-competitive antagonist at the N-methyl-D-aspartate (NMDA) type glutamate receptors. The rationale for this combination in OCD is to address both the serotonergic and glutamatergic systems, as glutamate dysregulation is implicated in the pathophysiology of the disorder. This specific treatment arm is part of the Phase 2 PROCEED trial led by the Shanghai Mental Health Center.
