Overview
Setanaxib + pembrolizumab is an investigational combination therapy for cancer, specifically evaluated in recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN). Setanaxib is a selective inhibitor of NADPH oxidase isoforms NOX1 and NOX4, which reduces oxidative stress and modulates fibroblast activity in the tumor microenvironment, showing antifibrotic and immunomodulatory effects. Pembrolizumab is a monoclonal antibody and immune checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor to restore antitumor immune responses. This combination leverages dual mechanisms: direct modulation of the tumor microenvironment and enhancement of antitumor immunity. Both drugs are administered together: setanaxib orally and pembrolizumab intravenously[1][3][5][7].
