Overview

SF1126 + nivolumab is an investigational combination therapy comprising SF1126, a dual inhibitor of phosphatidylinositol-3-kinase (PI3K, pan-isoform specific) and bromodomain-containing protein 4 (BRD4), and nivolumab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1). SF1126 disrupts key MYC-mediated oncogenic pathways, inhibiting tumor growth, while nivolumab blocks PD-1 to enhance T cell-mediated immune responses against cancer cells. This combination is being studied primarily for advanced or metastatic hepatocellular carcinoma (HCC), especially in patients with underlying liver disease, to enhance antitumor activity through both targeted molecular inhibition and immune checkpoint blockade[1].
