Drug intelligence / Profile preview
SFX-01 + fulvestrant is a **combination therapy** of two drugs: SFX-01, a stabilized synthetic formulation of sulforaphane (an isothiocyanate derivative from cruciferous vegetables), and fulvestrant, a selective estrogen receptor degrader (SERD). SFX-01 targets and inhibits the STAT3 signaling pathway and has demonstrated the ability to reduce cancer stem-like cells and reverse resistance to endocrine therapies in preclinical and clinical studies, particularly in estrogen receptor-positive (ER+) breast cancer. Fulvestrant acts by binding to and promoting degradation of the estrogen receptor, thereby inhibiting its signaling. The combination is primarily developed and investigated for metastatic ER+ breast cancer that has developed resistance to hormone therapies, with clinical evidence suggesting that SFX-01 can reduce fulvestrant-induced STAT3 activation and cancer stem cell activity, potentially enhancing the efficacy and overcoming endocrine resistance[1][3][5][6][7].
