Overview

**SGN-75 + everolimus** is a combination investigational therapy consisting of SGN-75, an antibody-drug conjugate (ADC), and everolimus, an oral mTOR inhibitor. SGN-75 is composed of an anti-CD70 monoclonal antibody linked to monomethyl auristatin F (MMAF), a cytotoxic agent, by proprietary technology developed by Seattle Genetics (now Seagen). SGN-75 selectively targets CD70-expressing tumor cells, delivering the potent cytotoxic agent directly to these cells to spare non-target tissues. Everolimus, marketed as Afinitor, inhibits the mammalian target of rapamycin (mTOR), thereby blocking cell growth, proliferation, and angiogenesis in cancer cells. The combination is under investigation for treatment of CD70-positive metastatic renal cell carcinoma, particularly in patients who have progressed after tyrosine kinase inhibitor therapy.
