Overview

**SHR-1316 + carboplatin + etoposide** is a combination regimen used primarily as a first-line therapy in patients with extensive stage small cell lung cancer (ES-SCLC). SHR-1316 is a recombinant fully humanized IgG4 monoclonal antibody targeting programmed death ligand 1 (PD-L1), an immune checkpoint molecule involved in suppression of anti-tumor immune responses. Carboplatin is a platinum-based chemotherapeutic that induces DNA cross-linking and apoptosis, while etoposide is a topoisomerase II inhibitor that leads to DNA breaks and cell death. This combination utilizes both immunotherapy (via immune checkpoint blockade) and cytotoxic chemotherapy for synergistic anti-tumor efficacy. SHR-1316 is developed by Hengrui Medicine; the combination has been under clinical investigation in studies such as the CAPSTONE-1 and NCT04562337 trials for ES-SCLC[1][7].
