Overview

SHR-1701 + albumin paclitaxel is a combination therapy used in oncology, currently under investigation primarily for pancreatic cancer. SHR-1701 is a bifunctional fusion protein composed of a monoclonal antibody against programmed death ligand 1 (PD-L1) fused to the extracellular domain of transforming growth factor-beta (TGF-β) receptor II, enabling simultaneous PD-L1 blockade and TGF-β sequestration. Albumin paclitaxel (nab-paclitaxel, Abraxane) is a solvent-free, albumin-bound formulation of the taxane chemotherapeutic paclitaxel, allowing for enhanced tumor delivery through albumin receptor-mediated transcytosis, improved pharmacokinetics, and reduced hypersensitivity reactions. The combination regimen is being clinically evaluated in untreated locally advanced or metastatic pancreatic adenocarcinoma, where SHR-1701 provides dual immune checkpoint and TGF-β pathway inhibition, and albumin paclitaxel arrests tumor cell division by stabilizing microtubules and blocking mitosis[3][6][2][5][8].
