SHR-3821 + fruquintinib

SHR-3821 + fruquintinib is a combination therapy being investigated for the treatment of advanced colorectal cancer (CRC). SHR-3821 is a humanized monoclonal antibody developed by Jiangsu Hengrui Pharmaceuticals that targets Transforming Growth Factor-beta (TGF-β), a cytokine involved in immune evasion, epithelial-mesenchymal transition, and fibrotic remodeling within the tumor microenvironment. Fruquintinib (brand name Elunate), developed by HUTCHMED, is a highly selective and potent oral small molecule inhibitor of Vascular Endothelial Growth Factor Receptors (VEGFR) 1, 2, and 3, which are critical drivers of tumor angiogenesis. This combination is currently being evaluated in an exploratory Phase II study led by investigator Meng Qiu at West China Hospital, targeting patients with advanced CRC who have failed standard therapies. The rationale for the combination is to simultaneously inhibit TGF-β-mediated immunosuppression and VEGFR-mediated blood vessel growth to overcome resistance in refractory colorectal tumors.
