Overview

SHR-A1811 + bevacizumab is a combination therapy under investigation for the treatment of HER2-positive breast cancer, including cases with brain metastases. SHR-A1811 is a third-generation antibody-drug conjugate (ADC) composed of an anti-HER2 antibody (trastuzumab), a cleavable linker, and a topoisomerase I inhibitor payload. It targets HER2-expressing or mutated tumors by delivering cytotoxic agents directly to cancer cells, resulting in cell death[5][7]. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing tumor blood supply[8]. The combination aims to enhance antitumor efficacy by simultaneously targeting tumor cells and their supporting vasculature. This regimen has shown promising activity in early-phase clinical trials for advanced breast cancer with brain metastasis[3].
