SHR-A1811 + capecitabine is a combination therapy under investigation for the treatment of unresectable or metastatic breast cancer with low HER2 expression. SHR-A1811 is an antibody-drug conjugate (ADC) composed of an anti-HER2 antibody (trastuzumab), a cleavable linker, and a topoisomerase I inhibitor payload. It targets HER2-expressing tumor cells, delivering the cytotoxic agent directly to these cells to induce cell death. Capecitabine is an oral prodrug of 5-fluorouracil (5-FU), classified as an antimetabolite, which interferes with DNA synthesis in rapidly dividing cancer cells. The combination aims to leverage the targeted cytotoxicity of SHR-A1811 and the systemic chemotherapeutic effect of capecitabine for enhanced efficacy in patients whose tumors express low levels of HER2[1][4][6].
