Overview

SHR-A1811 + everolimus is a combination therapy being investigated for the treatment of refractory triple-negative breast cancer (TNBC). SHR-A1811 is a next-generation antibody-drug conjugate (ADC) targeting Human Epidermal Growth Factor Receptor 2 (HER2), developed by Jiangsu Hengrui Pharmaceuticals. It consists of a humanized anti-HER2 monoclonal antibody conjugated to a potent topoisomerase I inhibitor payload (SHR152852) via a cleavable linker. Everolimus is an orally active inhibitor of the mechanistic target of rapamycin (mTOR), which plays a key role in cell growth, proliferation, and survival. The combination is being evaluated in a Phase II platform study led by Fudan University (NCT05834140) to explore precision therapy based on molecular subtyping in patients with refractory TNBC, specifically targeting those with HER2-low or HER2-positive expression and potential mTOR pathway activation.
