SHR-A1811 + pertuzumab is a combination of an investigational third-generation HER2-directed antibody-drug conjugate (ADC), SHR-A1811, and pertuzumab, a recombinant humanized monoclonal antibody targeting the HER2 protein. SHR-A1811 consists of an anti-HER2 antibody conjugated to a cytotoxic payload, designed for targeted delivery to HER2-overexpressing tumor cells, resulting in direct cytotoxicity to cancer cells. Pertuzumab binds to the extracellular dimerization domain (subdomain II) of HER2, inhibiting HER2 dimerization and downstream signaling that promotes cell growth and survival. This combination aims to improve clinical outcomes in patients with HER2-positive breast cancer, primarily as neoadjuvant therapy. Clinical development focuses on locally advanced or early-stage HER2-positive breast cancer, as assessed in phase II trials[1][2][3][5].
