Overview
**SHR-A1921 + cisplatin** is an investigational combination of a novel antibody-drug conjugate (ADC), SHR-A1921, with the platinum-based chemotherapy agent cisplatin. SHR-A1921 targets trophoblast cell-surface antigen 2 (TROP-2) and consists of a humanized anti–TROP-2 IgG1 monoclonal antibody linked via a tetrapeptide-based cleavable linker to a DNA topoisomerase I inhibitor payload. This ADC is being studied for its potential to treat advanced solid tumors, with a focus on ovarian and non-small cell lung cancers, including platinum-resistant and heavily pretreated disease. Cisplatin acts as a DNA crosslinking agent causing cytotoxicity in rapidly dividing cells. The combination is being evaluated for synergistic or additive anti-tumor activity in multiple ongoing clinical trials, including phase 1b/2 studies in advanced solid tumors.
