SI-B001 + SI-B003 is a combination of two investigational bispecific monoclonal antibodies for the treatment of advanced or metastatic solid tumors, particularly non-small cell lung cancer (NSCLC). SI-B001 (izalontamab) is a tetravalent bispecific antibody that targets the Epidermal Growth Factor Receptor (EGFR) and Human Epidermal Growth Factor Receptor 3 (HER3), inhibiting their oncogenic signaling and engaging immune effector mechanisms. SI-B003 (danvilostomig) is a tetravalent bispecific antibody targeting the immune checkpoints PD-1 and CTLA-4, restoring anti-tumor T cell activity through checkpoint blockade. Both antibodies are designed to work synergistically—SI-B001 directly targets tumor cell growth pathways, while SI-B003 enhances immune-mediated tumor destruction. The combination is in clinical development for patients with locally advanced or metastatic NSCLC who have failed prior therapies.[1][3][5][7]
