SIBP-03 + cetuximab is a combination therapy of two monoclonal antibodies: SIBP-03, a recombinant humanized anti-HER3 (ERBB3) IgG1 monoclonal antibody, and cetuximab, an anti-EGFR (epidermal growth factor receptor) monoclonal antibody. SIBP-03 inhibits HER3 activation (both ligand-dependent and ligand-independent) and downstream signaling via blocking heterodimerization with EGFR or HER2, disrupting the PI3K/AKT pathway that is crucial for tumor cell survival. It induces antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Cetuximab targets EGFR, blocking its signaling and proliferation functions. Preclinical and early clinical evidence supports that SIBP-03 synergizes with cetuximab, providing increased antitumor activity by concurrently inhibiting HER3 and EGFR pathways. The combination is under development for solid tumors, including squamous cell cancer, especially those with high EGFR/HER3 expression or activation[1][2][3][5][7].
