SIBP-03 + trastuzumab + docetaxel is a combination therapy consisting of three agents: - **SIBP-03:** a fully humanized monoclonal antibody targeting HER3 (ERBB3), developed to block HER3-mediated signaling, particularly the PI3K/AKT pathway. It exhibits potent antitumor effects and enhances the activity of other EGFR or HER2-targeted therapies, demonstrating potential synergy in preclinical models. - **Trastuzumab:** a well-established monoclonal antibody directed at HER2 (ERBB2), widely used in the treatment of HER2-positive cancers, disrupting signaling and mediating antibody-dependent cytotoxic effects. - **Docetaxel:** a cytotoxic chemotherapeutic agent interfering with microtubule function, leading to mitotic arrest and cell death, approved for various cancers including breast cancer. Preclinical data demonstrate that SIBP-03 significantly prolongs inhibition of HER3/PI3K/AKT signaling in xenograft models, especially when combined with HER2-targeting agents such as trastuzumab. Combination therapies leveraging these three drugs are under investigation, particularly for solid tumors and cancers expressing HER2 and HER3, with SIBP-03 currently in Phase I–II trials, including in China for squamous cell carcinoma and other solid tumors[1][3][5][7].
