Silver(I) 2,2'-bipyridine-triphenylphosphane is a mononuclear silver(I) coordination complex being investigated for its potent anticancer activity, particularly in models of ovarian and colorectal cancer. The complex consists of a silver(I) center coordinated by a bidentate 2,2'-bipyridine ligand and a triphenylphosphane ligand, resulting in a distorted trigonal planar geometry. Its primary mechanism of action is the induction of paraptosis, a non-apoptotic form of programmed cell death characterized by endoplasmic reticulum stress and the formation of large cytoplasmic vacuoles. This mechanism allows the compound to bypass common resistance pathways associated with platinum-based chemotherapies, such as carboplatin and oxaliplatin resistance or p53 deletions. The research is driven by an academic consortium including the Ruđer Bošković Institute, the Medical University of Vienna, and the University of Lisbon.
