Overview

Silver + glutamine + thiosemicarbazone (Ag@Gln-TSC NPs) is a research-stage nanoparticle-based therapeutic candidate described in a 2024 academic study for the treatment of colon cancer. The compound consists of silver nanoparticles functionalized with glutamine to improve biocompatibility and cellular internalization, and conjugated with thiosemicarbazide to impart anticancer activity. In preclinical models using the SW480 colon cancer cell line, Ag@Gln-TSC NPs demonstrated potent cytotoxicity, inducing apoptosis in approximately 85.9% of treated cells and causing S-phase cell cycle arrest. Its mechanism of action involves the activation of Caspase-3 and the upregulation of the CASP8 gene, alongside the downregulation of the HULC long non-coding RNA and the PPFIA4 oncogene. Additionally, the thiosemicarbazone component is proposed to disrupt iron acquisition mechanisms within the tumor microenvironment.
