SIM0417 (also known as simnotrelvir or SSD8432) is an orally administered small-molecule inhibitor targeting the coronavirus main protease (3CLpro), developed for the treatment of COVID-19. Ritonavir is co-administered as a pharmacokinetic enhancer to inhibit CYP3A-mediated metabolism and increase systemic exposure of SIM0417. Midazolam is a short-acting benzodiazepine and a sensitive substrate of CYP3A4, often used in drug-drug interaction studies to assess CYP3A inhibition. The combination "SIM0417 + ritonavir + midazolam" has been studied in healthy volunteers to evaluate pharmacokinetic interactions; co-administration with ritonavir significantly increases plasma concentrations of midazolam due to strong inhibition of CYP3A4 by ritonavir and weak inhibition by simnotrelvir[1][2][5]. This combination is not intended for therapeutic use but rather for clinical pharmacology research on drug-drug interactions.
