SIM0501 is an antibody-drug conjugate (ADC) targeting Receptor tyrosine kinase-like orphan receptor 1 (ROR1), developed by Simcere Pharmaceutical (specifically its subsidiary Shandong Simcere Medgenn). The drug consists of a humanized anti-ROR1 monoclonal antibody conjugated to a potent cytotoxic payload, typically a topoisomerase I inhibitor, via a cleavable linker. ROR1 is an oncofetal protein that is highly expressed in a variety of malignancies—including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and various B-cell hematological malignancies—while remaining largely absent in healthy adult tissues. By specifically binding to ROR1, SIM0501 delivers its cytotoxic payload directly into tumor cells through receptor-mediated endocytosis, aiming to maximize anti-tumor efficacy while minimizing systemic toxicity. It is currently being evaluated in Phase 1 clinical trials for advanced solid tumors and hematological malignancies.
