Simeprevir + daclatasvir is an oral combination regimen of two direct-acting antiviral agents used for the treatment of chronic hepatitis C virus (HCV) infection. Simeprevir is an HCV NS3/4A protease inhibitor that blocks viral replication by inhibiting the cleavage of the HCV polyprotein. Daclatasvir is an HCV NS5A inhibitor that interferes with viral RNA replication and virion assembly. This combination has been studied primarily in patients with genotype 1b infection and advanced liver disease, showing strong antiviral activity and a high rate of sustained virologic response (SVR12), though some risk of viral breakthrough exists[6][8]. Both drugs are administered orally once daily.
