**Simmitecan + thalidomide** is a combination regimen under clinical investigation for advanced solid tumors, primarily colorectal cancer. Simmitecan is a novel, potent lipophilic camptothecin derivative that acts as an inhibitor of **topoisomerase I**, resulting in anti-tumor activity by interfering with DNA replication and transcription in cancer cells.
