Overview

Simmitinib + irinotecan liposome is a combination therapy consisting of simmitinib (details on this agent are not available in the provided sources or current literature) and irinotecan liposome. Irinotecan liposome is a nanoliposomal formulation of the topoisomerase I inhibitor irinotecan, encapsulated within a lipid bilayer vesicle. This formulation prolongs circulation time and enhances tumor accumulation via the enhanced permeability and retention effect. Irinotecan and its active metabolite SN-38 inhibit topoisomerase I by binding to the topoisomerase I–DNA complex, preventing repair of single-strand DNA breaks during replication, leading to double-strand DNA damage and cell death. The primary indication for irinotecan liposome (Onivyde) is metastatic pancreatic cancer, typically in combination with other agents such as fluorouracil and leucovorin[1][3][4][7]. The developer/manufacturer for Onivyde is Ipsen Biopharmaceuticals[5][7].
