SIM0417 + ritonavir + rifampicin is a drug combination used in clinical pharmacology studies to evaluate drug-drug interactions (DDI). It consists of **simnotrelvir** (SIM0417), an oral small-molecule antiviral that targets the SARS-CoV-2 3-chymotrypsin-like protease (3CLpro), which is essential for viral replication; **ritonavir**, a pharmacokinetic booster that inhibits the CYP3A4 enzyme to increase the plasma exposure of simnotrelvir; and **rifampicin**, a potent CYP3A4 inducer used as a clinical probe to assess the impact of metabolic induction on the pharmacokinetics of the simnotrelvir/ritonavir regimen. Simnotrelvir was developed by Simcere Pharmaceutical in collaboration with the Shanghai Institute of Materia Medica and the Wuhan Institute of Virology. The co-packaged product of simnotrelvir and ritonavir is marketed as Xiannuoxin for the treatment of mild-to-moderate COVID-19.
