Simvastatin + interferon beta-1a is a combination of a cholesterol-lowering drug of the statin class (simvastatin) and an immunomodulatory protein (interferon beta-1a) used principally in research for the treatment of relapsing-remitting multiple sclerosis. Simvastatin acts as an inhibitor of HMG-CoA reductase, thereby reducing cholesterol synthesis and exerting anti-inflammatory effects. Interferon beta-1a is a recombinant type I interferon that binds to interferon receptors on cell surfaces and modulates immune activity principally by altering cytokine expression and reducing lymphocyte activation. The combination has been evaluated for potential synergistic or additive effects in reducing disease activity in multiple sclerosis, but high-quality evidence from the SIMCOMBIN trial shows the combination does not provide benefit beyond interferon beta-1a alone and may display pharmacological antagonism.
