Simvastatin + zoledronic acid is a combination therapy that creates a sequential blockade of the mevalonate pathway. Simvastatin inhibits HMG-CoA reductase, while zoledronic acid inhibits farnesyl pyrophosphate synthase. This combination has shown synergistic effects in inducing apoptosis in cancer cells, suppressing Dickkopf-1 in breast cancer, and treating parasitic infections like toxoplasmosis. The synergy allows for lower concentrations of each drug to be used while maintaining or enhancing efficacy.
