Overview

Sinibolimab + lenvatinib is a combination therapy regimen being evaluated for the neoadjuvant treatment of locally advanced unresectable differentiated thyroid carcinoma. Sinibolimab is an anti-PD-1 monoclonal antibody that functions as an immune checkpoint inhibitor, enhancing the immune system's ability to detect and destroy tumor cells by blocking the interaction between PD-1 and its ligands. Lenvatinib is an oral multikinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1-3), fibroblast growth factor receptors (FGFR1-4), RET, KIT, and platelet-derived growth factor receptor alpha (PDGFRα), thereby providing anti-angiogenic and anti-proliferative effects. This combination is currently being studied in a Phase II clinical trial led by Zhejiang University to assess its efficacy in reducing tumor burden and making unresectable tumors eligible for surgical resection (R0/R1 resection).
