Drug intelligence / Profile preview
**Sintilimab + anlotinib + chemotherapy** is an investigational combination cancer regimen that pairs the PD-1 immune checkpoint inhibitor sintilimab with the oral multitarget tyrosine kinase inhibitor anlotinib and a cytotoxic chemotherapy backbone. Sintilimab is a fully human IgG4 monoclonal antibody that blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell-mediated antitumor immunity. Anlotinib is a small-molecule inhibitor targeting multiple receptor tyrosine kinases, including VEGFR1/2/3, FGFR1-4, PDGFR-alpha/beta, and c-Kit, which suppresses tumor angiogenesis and cell proliferation. The chemotherapy backbone varies depending on the clinical study and tumor type (e.g., nab-paclitaxel, gemcitabine/cisplatin, or albumin paclitaxel plus nedaplatin) to directly induce tumor cell death. This triplet regimen is being evaluated in Phase 2 clinical trials for various solid tumors, including non-small cell lung cancer (NSCLC), extensive-stage small cell lung cancer (ES-SCLC), biliary tract cancer (BTC), esophageal cancer, and triple-negative breast cancer (TNBC), demonstrating promising synergistic efficacy and manageable safety profiles.
