Sintilimab + AVD is an investigational combination regimen consisting of the anti-PD-1 monoclonal antibody sintilimab and the chemotherapy backbone AVD (doxorubicin, vinblastine, and dacarbazine). Sintilimab is a fully humanized IgG4 monoclonal antibody that binds to the programmed death-1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. AVD is a standard chemotherapy regimen for Hodgkin lymphoma that omits bleomycin to reduce pulmonary toxicity. This combination is being evaluated in a phase II clinical trial for patients with newly diagnosed unfavorable stage II-IV classical Hodgkin lymphoma (cHL) who show a suboptimal response (Deauville score 4) to initial induction chemotherapy, aiming to provide a more effective first-line treatment and potentially avoid high-dose chemotherapy and radiotherapy.
