Drug intelligence / Profile preview
This is a combination therapy comprising **sintilimab** (a PD-1 inhibitor monoclonal antibody), **bevacizumab** (a VEGF inhibitor monoclonal antibody), and **tafolecimab** (a PCSK9 inhibitor monoclonal antibody). - **Sintilimab** (TYVYT) is a fully human IgG4 anti-PD-1 monoclonal antibody that blocks the interaction between PD-1 and its ligands, thereby reactivating T-cells to attack tumor cells. - **Bevacizumab** (BYVASDA) is a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), suppressing angiogenesis in tumors. - **Tafolecimab** (SINTBILO) is a fully human IgG2 monoclonal antibody targeting proprotein convertase subtilisin/kexin type 9 (PCSK9), reducing LDL-C by blocking PCSK9-mediated LDL receptor degradation. This triple regimen is being evaluated as an immuno-oncology and anti-angiogenic/metabolic therapy in solid tumors, especially in patients with previous anti-PD-1/PD-L1 exposure. Tafolecimab is also approved for hypercholesterolemia but is being tested here for possible immune-modulatory effects in cancer[2][5].
