Overview

This investigational combination therapy consists of the anti-PD-1 monoclonal antibody sintilimab, the platinum-based chemotherapeutic agent carboplatin, and the albumin-bound taxane nab-paclitaxel. It is currently being evaluated in a Phase II clinical trial led by Sun Yat-Sen Memorial Hospital of Sun Yat-Sen University as a neoadjuvant (pre-surgical) treatment for patients with resectable salivary gland malignant neoplasms. Sintilimab works by blocking the interaction between the Programmed cell death protein 1 (PD-1) receptor on T-cells and its ligands (PD-L1/PD-L2) on tumor cells, thereby restoring the immune system's ability to identify and destroy cancer cells. Carboplatin acts as a DNA alkylating agent that induces interstrand and intrastrand DNA cross-links, inhibiting DNA synthesis and triggering apoptosis. Nab-paclitaxel is a nanoparticle formulation of paclitaxel that stabilizes microtubules, preventing their disassembly and arresting the cell cycle. The combination aims to leverage the synergistic potential of immune checkpoint inhibition and cytotoxic chemotherapy to improve pathological response rates and surgical outcomes in salivary gland cancer.
