Overview

Sintilimab is a recombinant human monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells. By binding to PD-1, it blocks its interaction with the ligands PD-L1 and PD-L2, thereby inhibiting immune checkpoint signaling and restoring T-cell-mediated immune responses against tumor cells. When combined with chemotherapy agents (such as pemetrexed and platinum-based regimens), this combination has demonstrated improved efficacy in treating advanced cancers—particularly non-small cell lung cancer (NSCLC)—by increasing overall response rates, prolonging progression-free survival (PFS), and overall survival (OS) compared to chemotherapy alone. Sintilimab is developed by Innovent Biologics in partnership with Eli Lilly and marketed under brand names such as Tyvyt in China. It has been approved for several indications including NSCLC, hepatocellular carcinoma, esophageal squamous cell carcinoma, and classical Hodgkin lymphoma[1][3][4][5][6][8].
