This is a multi-agent combination regimen consisting of five drugs used in the treatment of advanced or metastatic non-small cell lung cancer (NSCLC) and potentially other solid tumors. The regimen includes: - **Sintilimab**: a fully human IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1/PD-L2 to enhance T-cell mediated anti-tumor immunity. - **Cisplatin** and **Carboplatin**: platinum-based chemotherapeutic agents that form DNA crosslinks, leading to apoptosis in rapidly dividing tumor cells. - **Pemetrexed**: a multitargeted antifolate that inhibits several folate-dependent enzymes involved in nucleotide synthesis, thereby disrupting DNA replication and cell division. - **Gemcitabine**: a nucleoside analog that incorporates into DNA during replication, causing chain termination and apoptosis. This combination leverages both immunotherapy (sintilimab) and cytotoxic chemotherapy (the other four agents), aiming for synergistic anti-tumor effects. It is primarily investigated for use as first-line therapy or subsequent lines in advanced NSCLC, especially nonsquamous subtypes[4][6][7]. Sintilimab was developed by Innovent Biologics and Eli Lilly[6][9].
