IBI308 + cisplatin + pemetrexed is a combination chemo-immunotherapy regimen consisting of sintilimab (IBI308), a recombinant fully human anti-PD-1 monoclonal antibody, and the chemotherapy agents cisplatin and pemetrexed. Sintilimab, developed by Innovent Biologics, restores anti-tumor immune responses by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2). Cisplatin is a platinum-based agent that induces DNA cross-linking, thereby inhibiting DNA replication and transcription. Pemetrexed is a folate antimetabolite that inhibits multiple enzymes involved in pyrimidine and purine synthesis, including thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT). This combination has demonstrated efficacy in Phase 3 trials (such as ORIENT-31) for patients with EGFR-mutated nonsquamous non-small cell lung cancer (NSCLC) who have progressed on EGFR TKI therapy, and has been evaluated in Phase 1 studies for various other advanced solid tumors.
