**Sintilimab + gemcitabine + cisplatin** is a combination regimen used primarily in oncology as a first-line treatment for several advanced or metastatic cancers, notably biliary tract cancer (BTC), non-small cell lung cancer (NSCLC), and muscle-invasive bladder cancer (MIBC). Sintilimab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), thereby inhibiting the PD-1/PD-L1 interaction and restoring antitumor T cell activity (checkpoint inhibition). Gemcitabine and cisplatin are cytotoxic chemotherapeutic agents: gemcitabine is a nucleoside analog inhibiting DNA synthesis, and cisplatin is a platinum-based DNA crosslinker inducing apoptosis. The combination leverages immunotherapy with cytotoxic chemotherapy for synergistic antitumor efficacy, and has demonstrated improved progression-free survival and acceptable safety profiles in phase II and phase III trials in oncologic indications including advanced BTC, locally advanced/metastatic NSCLC, and as neoadjuvant therapy in MIBC[1][2][3][5][8].
