sintilimab + IBI310 + chemotherapy

This combination therapy regimen consists of sintilimab (an anti-PD-1 monoclonal antibody), IBI310 (an anti-CTLA-4 monoclonal antibody), and chemotherapy (specifically pemetrexed/carboplatin or nab-paclitaxel/carboplatin). It is currently being evaluated in a Phase II neoadjuvant trial led by Shanghai Pulmonary Hospital for patients with resectable, PD-L1-negative stage II-IIIB non-small cell lung cancer (NSCLC). Sintilimab, developed by Innovent Biologics, blocks the PD-1/PD-L1 pathway to restore T-cell activity, while IBI310, also from Innovent, targets CTLA-4 to enhance T-cell priming and proliferation. The addition of chemotherapy is intended to provide direct cytotoxic effects and potentially increase the release of tumor antigens, thereby enhancing the efficacy of the dual checkpoint blockade in patients who typically show poor responses to single-agent immunotherapy.
