Overview

This combination regimen consists of sintilimab, an anti-PD-1 monoclonal antibody; IBI310, an anti-CTLA-4 monoclonal antibody; and lenvatinib, a multi-kinase inhibitor. It is currently being evaluated in a Phase 2 clinical trial (NCT05534282) sponsored by Peking University for the treatment of non-metastatic gastric or colon cancers that are mismatch repair deficient (dMMR) or microsatellite instability-high (MSI-H). The therapeutic strategy aims for organ preservation by utilizing an initial phase of PD-1/PD-L1 antibody monotherapy followed by an intensive combination of the three agents to achieve a complete clinical response, thereby potentially avoiding the need for radical surgery. Sintilimab and IBI310 are developed by Innovent Biologics, while lenvatinib is developed by Eisai.
