A combination therapy regimen consisting of the PD-1 inhibitor sintilimab, the multi-kinase inhibitor lenvatinib, and the albumin-bound chemotherapy agent nab-paclitaxel. This triplet combination is being investigated in clinical trials, notably by Zhejiang Cancer Hospital, as a first-line treatment for patients with recurrent or metastatic triple-negative breast cancer (TNBC). Sintilimab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor to enhance anti-tumor immune responses. Lenvatinib is a small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1-3), fibroblast growth factor receptors (FGFR1-4), and other kinases to inhibit angiogenesis and tumor growth. Nab-paclitaxel is a nanoparticle formulation of paclitaxel that stabilizes microtubules, leading to cell cycle arrest and apoptosis. The combination aims to provide a multi-pronged attack on TNBC by combining immune checkpoint blockade, anti-angiogenesis, and cytotoxic chemotherapy.
