This combination regimen consists of sintilimab (also known as sindilizumab), a recombinant humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, in combination with paclitaxel polymeric micelles and platinum-based chemotherapy (cisplatin or carboplatin). Sintilimab works as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell mediated anti-tumor activity. Paclitaxel polymeric micelles represent a nanoparticle formulation of the taxane paclitaxel, designed to improve solubility and delivery while disrupting microtubule dynamics to induce cell cycle arrest. The platinum agents (cisplatin or carboplatin) act as DNA alkylators, forming cross-links that lead to apoptosis. This specific chemo-immunotherapy combination is being evaluated by the Jiangsu Cancer Institute & Hospital in a Phase 2 clinical trial (NCT05782426) as a first-line treatment for patients with advanced or metastatic non-squamous non-small cell lung cancer (NSCLC) who do not harbor EGFR mutations or ALK rearrangements.
