The combination of sintilimab and pegaspargase is an investigational regimen for the treatment of advanced or early-stage extranodal natural killer/T-cell lymphoma (ENKTL/NKTCL). Sintilimab is a fully human anti-PD-1 monoclonal antibody that blocks the PD-1/PD-L1 immune checkpoint, thereby enhancing T cell-mediated antitumor immunity. Pegaspargase is an anti-metabolic agent, a modified form of L-asparaginase, which depletes asparagine in tumor cells, leading to selective cytotoxicity in lymphoblasts and certain lymphoma cells. This chemo-free regimen has demonstrated promising efficacy and safety in clinical trials for newly diagnosed advanced stage NKTCL[6][9].
