**sintilimab + pyrotinib** is a combination therapy under clinical investigation, particularly for HER2-positive malignancies. **Sintilimab** is a fully human anti-programmed death 1 (PD-1) monoclonal antibody that blocks the PD-1/PD-L1 pathway, restoring T-cell activity for immune-mediated tumor killing. **Pyrotinib** is an oral, irreversible tyrosine kinase inhibitor that targets both human epidermal growth factor receptor 2 (HER2) and EGFR pathways, inhibiting cancer cell proliferation. The combination is being studied for synergistic anti-tumor efficacy in HER2-positive cancers, such as advanced breast cancer and non-small-cell lung cancer (NSCLC)[4][8]. Both agents are products of Chinese pharmaceutical innovation, with sintilimab developed by Innovent Biologics and Eli Lilly, and pyrotinib by Jiangsu Hengrui Medicine. Primary indications include HER2-positive breast cancer and HER2-mutated NSCLC.
