Sintilimab + ramucirumab is an investigational combination therapy for advanced or metastatic gastric and gastroesophageal junction cancers. Sintilimab is a recombinant human IgG4 monoclonal antibody that targets and inhibits programmed cell death protein 1 (PD-1), thereby blocking immune checkpoint signaling and enhancing antitumor immune responses. Ramucirumab is a human IgG1 monoclonal antibody that binds vascular endothelial growth factor receptor 2 (VEGFR-2), blocking the binding of VEGF ligands (VEGF-A, VEGF-C, and VEGF-D) and preventing tumor angiogenesis. Together, the combination aims to synergistically inhibit immune evasion and angiogenic signaling, improving overall survival and response rates for these malignancies[2][6][8][9].
