Overview

Sintilimab + ramucirumab + paclitaxel is an investigational triple-combination regimen being evaluated for the treatment of advanced gastric cancer. The regimen consists of three distinct therapeutic agents: **sintilimab** (Tyvyt), a recombinant fully human IgG4 monoclonal antibody that targets the PD-1 immune checkpoint to restore anti-tumor T-cell activity; **ramucirumab** (Cyramza), a human IgG1 monoclonal antibody that acts as a VEGFR-2 antagonist to inhibit tumor angiogenesis; and **paclitaxel**, a taxane-class small molecule chemotherapy that stabilizes microtubules to arrest cell division. This combination is primarily being studied in Phase 2 clinical trials (e.g., NCT06564298) for patients who have experienced short-term recurrence after postoperative adjuvant therapy, with the goal of leveraging the synergistic effects of immune checkpoint blockade, anti-angiogenic therapy, and cytotoxic chemotherapy.
