Overview

Sintilimab + surufatinib is an investigational combination therapy for solid tumors, particularly being studied in advanced and metastatic cancers such as pancreatic cancer, gastric cancer/gastroesophageal junction cancer (GC/GEJC), and neuroendocrine neoplasms. Sintilimab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Surufatinib is a small-molecule multi-targeted kinase inhibitor that selectively inhibits vascular endothelial growth factor receptors 1–3 (VEGFR1–3), fibroblast growth factor receptor 1 (FGFR1), and colony-stimulating factor 1 receptor (CSF1R). This dual approach aims to both modulate the tumor microenvironment by inhibiting angiogenesis and tumor-associated macrophages, while also promoting T-cell mediated anti-tumor activity through PD-1 blockade. The combination has shown preliminary efficacy with manageable toxicity in early-phase clinical trials for several solid tumors[1][2][4][5].
