Overview

Sintilimab + tolecimab is a combination therapy comprising the programmed cell death protein 1 (PD-1) inhibitor sintilimab and the proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor tolecimab. Sintilimab is a humanized IgG4 monoclonal antibody that restores anti-tumor T-cell activity by blocking the interaction between PD-1 and its ligands, PD-L1 and PD-L2. Tolecimab is a humanized monoclonal antibody that targets PCSK9, a protein primarily known for regulating cholesterol levels by promoting the degradation of LDL receptors. In the context of oncology, PCSK9 inhibition is being investigated for its potential to upregulate MHC-I expression on tumor cells, thereby enhancing their visibility to the immune system and potentially overcoming resistance to immune checkpoint inhibitors. This combination is currently being evaluated in Phase II clinical trials for patients with advanced driver gene-negative non-small cell lung cancer (NSCLC) who have high PCSK9 expression and have failed prior first-line immunotherapy.
