Apatolimab + tovolimab is an investigational combination immunotherapy regimen developed by Innovent Biologics, consisting of two monoclonal antibodies: **sintilimab** (also known as apatolimab or IBI308) and **tovolimab** (IBI310). Sintilimab is a recombinant humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. Tovolimab is a human monoclonal antibody targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), a checkpoint molecule that downregulates T-cell activation. By combining these two agents, the regimen achieves dual checkpoint blockade, potentially overcoming resistance mechanisms associated with single-agent PD-1 inhibition. The combination is being evaluated in various clinical trials, including Phase 2 and Phase 3 studies for solid tumors such as gastric adenocarcinoma, gastroesophageal junction adenocarcinoma, and melanoma, often in combination with anti-angiogenic agents or chemotherapy.
