Overview

This combination therapy is an investigational chemo-immunotherapy and targeted therapy regimen primarily being evaluated for the treatment of advanced non-small cell lung cancer (NSCLC), particularly in patients with EGFR mutations who have progressed on prior EGFR tyrosine kinase inhibitor (TKI) therapy. The regimen consists of **sintilimab**, a recombinant humanized IgG4 monoclonal antibody that acts as a checkpoint inhibitor by binding to the programmed cell death protein 1 (PD-1) receptor; **vormetinib** (also known as furmonertinib), a third-generation, irreversible EGFR-TKI designed to selectively inhibit both EGFR sensitizing mutations and the T790M resistance mutation; and **platinum-based chemotherapy** (typically a doublet of pemetrexed and carboplatin or cisplatin). The combination aims to overcome tumor heterogeneity and resistance by simultaneously targeting the EGFR signaling pathway, enhancing the anti-tumor immune response, and inducing direct cytotoxic DNA damage. This specific triplet is being studied in investigator-initiated trials led by institutions such as the Second Xiangya Hospital of Central South University.
