Overview

Siplizumab + belatacept is an investigational combination therapy comprising siplizumab (TCD601), a humanized monoclonal antibody targeting CD2, and belatacept, a selective T-cell costimulation blocker that inhibits CD28-mediated T-cell activation by binding CD80 and CD86. This combination is under clinical evaluation as a calcineurin-inhibitor–free immunosuppressive regimen for the prevention of rejection in de novo renal transplant recipients. Siplizumab depletes or modulates T cells via CD2 signaling, while belatacept provides co-stimulation blockade, resulting in synergistic immunosuppression to prevent allograft rejection[1][3][5][7].
