Overview

**Siplizumab + cyclophosphamide** is a combination of a humanized monoclonal antibody and a cytotoxic alkylating agent used experimentally in the treatment of cancer and for immunosuppressive protocols, including transplantation. Siplizumab is an IgG1κ monoclonal antibody targeting the CD2 receptor on T-lymphocytes and natural killer (NK) cells, inhibiting T-cell activation, expansion, and leading to depletion of T and NK cells[3][6]. Cyclophosphamide is a small molecule alkylating agent that crosslinks DNA, causing cell death, particularly in rapidly dividing cells. This combination has mainly been tested in phase I studies for aggressive peripheral T-cell lymphomas, often as part of a multi-agent chemotherapy regimen, and in protocols for tolerance induction in transplantation, where cyclophosphamide is used for non-myeloablative conditioning and siplizumab aims to modulate immune activation and promote tolerance[5][3][6].
