Sipuleucel-T + leuprolide acetate is a combination therapy where sipuleucel-T is an autologous cellular immunotherapy (cancer vaccine) indicated for metastatic castration-resistant prostate cancer (mCRPC), and leuprolide acetate is a synthetic gonadotropin-releasing hormone (GnRH/LHRH) agonist used for androgen deprivation. Sipuleucel-T activates patient-derived antigen-presenting cells with a recombinant fusion protein (PA2024; prostatic acid phosphatase linked to GM-CSF) to provoke a T-cell immune response targeted against prostate cancer cells expressing PAP. Leuprolide acetate induces testosterone suppression by downregulating the pituitary-gonadal axis, assisting in the underlying hormone-refractory disease setting[1][2][3].
